information for you.

  • Keep this leaflet. You may need to read it again.
  • If you have any questions, ask your doctor, pharmacist or nurse.
  • This medicine has been prescribed for you only. Do not pass it on to others, even if their symptoms are the same as yours, as it could be harmful.
  • If you get any side effects, even those not listed in this leaflet, tell your doctor, pharmacist or nurse. See section 4.

1. What is Lyxumia and what is it used for

Lyxumia contains the active substance lixisenatide.
It is an injectable medicine that helps your body control the level of sugars in the
blood when it is too high. It is used in adults with type 2 diabetes.
Lyxumia is used with other medicines for diabetes when these are not sufficient to control the
blood sugar levels, such as:

  • oral antidiabetic drugs (such as metformin, pioglitazone, sulfonylureas) and/or
  • a basal insulin, a type of insulin that works all day long.

2. What you need to know before using Lyxumia

Do not use Lyxumia

  • if you are allergic to lixisenatide or any of the other ingredients of this medicine (listed in section 6).

Warnings and precautions
Talk to your doctor, pharmacist or nurse before using Lyxumia:

  • if you have type 1 diabetes or “diabetic ketoacidosis” (a complication of diabetes that occurs when the body cannot convert glucose because there is not enough insulin) as this medicine is not appropriate for you
  • if you have or have had inflammation of the pancreas (pancreatitis)
  • if you have a serious problem with your stomach or intestines, such as a disease of the stomach muscles called “gastroparesis”, which causes delayed stomach emptying
  • if you have a serious kidney disease or are on dialysis; in these cases the use of this medicine is not recommended
  • if you are also taking a sulfonylurea or a basal insulin. In this case, a decrease in blood sugar (hypoglycemia) may occur. Your doctor may check your blood sugar level and then decide to reduce your dose of basal insulin or sulfonylurea. Lyxumia should not be used in combination with a basal insulin together with a sulfonylurea.
  • if you are taking other medicines, as there are other medicines such as antibiotics or gastro-resistant tablets or capsules that should not remain in the stomach for a long time (see Other medicines and Lyxumia)
  • if you experience fluid loss/dehydration, e.g. in case of vomiting, nausea and diarrhoea. It is important to avoid dehydration by drinking plenty of fluids, especially at the beginning of treatment with Lyxumia.
  • if you suffer from heart problems that can cause shortness of breath or swelling of the ankles, as there is limited experience in this population

Children and adolescents
There is no data on Lyxumia in children and adolescents under 18 years of age, therefore the use of Lyxumia
in this age group is not recommended.

Other medicines and Lyxumia
Tell your doctor, pharmacist or nurse if you are taking, have recently taken or might take
any other medicines.
Lyxumia can affect the effects of some medicines taken by mouth. It is possible that you
need to take some medicines, such as antibiotics, gastro-resistant tablets or capsules that
should not remain in the stomach for too long at least one hour before or four hours after the injection
of Lyxumia.

Pregnancy and breastfeeding
Lyxumia should not be used during pregnancy. It is not known whether Lyxumia can be harmful to
the fetus.
Lyxumia should not be used during breastfeeding. It is not known whether Lyxumia passes into breast milk.
If you are pregnant, suspect you are pregnant, or are planning to become pregnant, or if you are breastfeeding
ask your doctor or pharmacist for advice before using this medicine.

Driving and using machines
If you use Lyxumia with a sulfonylurea or a basal insulin, you may experience a decrease in blood
sugar (hypoglycemia). This may make it difficult to concentrate and may cause dizziness or
drowsiness. If this happens, do not drive vehicles or use tools or machinery.

Important information about some ingredients of Lyxumia
This medicine contains less than 1 mmol of sodium per dose, i.e. it is practically "sodium-free".
This medicine contains metacresol, which can cause allergic reactions.

3. How to use Lyxumia

Use this medicine following the instructions of your doctor, pharmacist or nurse exactly. If you have any doubts, ask your doctor, pharmacist or nurse.

How much medicine to inject

  • The initial dose is 10 micrograms once daily for the first 14 days - injected with the greenpen.
  • From that point on, the dose will be 20 micrograms once daily - using the purplepen.

When to inject the medicine
Inject Lyxumia within one hour before any meal of the day. Preferably inject Lyxumia every day before the same meal, once you have chosen the most convenient meal for the injection.

Where to inject the medicine
Inject Lyxumia into the skin (subcutaneously) in the abdominal area, the upper part of the thigh or the upper part of the arm.

How to use the pre-filled pens
Before using the pre-filled pen for the first time, your doctor or nurse will show you how to inject Lyxumia.

  • Always read the “Instructions for Use” contained in the package.
  • Always use the pen as described in the Instructions for Use.

Other important information about using the pre-filled pens
The “Instructions for Use” contain further information on using the pens. The most important points are as follows:

  • Always use a new needle for each injection. Dispose of the needles after each use, in a sharps container in accordance with local regulations. Ask your doctor, nurse or pharmacist for information on how to dispose of sharps.
  • Use only needles compatible with the Lyxumia pen (see “Instructions for Use”).
  • Before using it for the first time, you must activate the Lyxumia pen.This will ensure that it works correctly and that the dose of the first injection is correct.
  • If you believe that the Lyxumia pen may be damaged, do not use it. Take a new one. Do not try to repair the pen.

If you use more Lyxumia than you should
If you use more Lyxumia than you should, contact your doctor immediately. A dose that is too high of Lyxumia can cause you to feel unwell or make you feel sick.

If you forget to use Lyxumia
If you miss a dose of Lyxumia, you can inject it within one hour before your next meal. Do not take two doses in the same day to make up for a missed injection.

If you stop using Lyxumia
Do not stop using Lyxumia without first talking to your doctor. If you stop using Lyxumia, your blood sugar levels may increase.
If you have any further questions on the use of this medicine, ask your doctor, pharmacist or nurse.

4. Possible side effects

Like all medicines, this medicine can cause side effects, although not everybody gets them.
In patients treated with Lyxumia, some serious allergic reactions (such as anaphylaxis) have been reported rarely.
If you experience symptoms such as swelling of the face, tongue or throat that make it difficult to breathe, seek immediate medical attention.
Stop taking Lyxumia and contact your doctor immediately if you notice the onset of the
following serious side effects:

  • Severe and persistent abdominal pain (stomach area), which may extend to the back, as well as nausea and vomiting, which may be a sign of inflammation of the pancreas (pancreatitis).

The most common side effects reported with Lyxumia that may affect more than 1 in 10 users
are nausea (feeling unwell) and vomiting. These side effects are usually mild and generally disappear over time.
Other side effects include:

  • Diarrhea
  • Headache
  • Low blood sugar (hypoglycemia), especially when Lyxumia is used with insulin or a sulfonylurea. Warning signs of low blood sugar can include cold sweat, pale and cold skin, headache, feeling sleepy, weakness, dizziness, confusion or irritability, hunger, fast heartbeat and nervousness. Your doctor will tell you what to do if you experience low blood sugar. It is more likely to happen if you are also taking a sulfonylurea or a basal insulin. Your doctor may reduce the dose of these medicines before you start using Lyxumia.

Common side effects: may affect up to 1 in 10 people

  • Influenza
  • Common cold (upper respiratory tract infection)
  • Dizziness
  • Indigestion (dyspepsia)
  • Back pain
  • Cystitis
  • Viral infection
  • Low blood sugar levels (when Lyxumia is taken with metformin)
  • Somnolence
  • Reactions at the injection site (such as itching)

Uncommon side effects: may affect up to 1 in 100 people

  • Urticaria
  • Gallstones
  • Inflamed gallbladder

Rare side effect (may affect up to one in 1000 people)

  • A delay in stomach emptying

Reporting of side effects
If you get any side effects, talk to your doctor, pharmacist or nurse.
You can also report side effects directly through the national reporting system listed in Annex V.
By reporting side effects you can help provide more information on the safety
of this medicine.

5. How to store Lyxumia

Keep this medicine out of the sight and reach of children.
Do not use this medicine after the expiry date which is stated on the pen label and on the
pack after Exp./EXP. The expiry date refers to the last day of that month.
Before first use
Store in a refrigerator (2 °C – 8 °C). Do not freeze. Keep away from the freezer compartment.
During pen use
The pen can be used for 14 days if stored at a temperature below 30°C. Do not
freeze. Do not store with the needle attached. When you are not using the pen, always replace the cap
to protect it from light.
Do not dispose of any medicine in wastewater or household waste. Ask your pharmacist how to
dispose of medicines you no longer use. This will help protect the environment.

6. Content of the pack and other information

What Lyxumia contains

  • The active substance is lixisenatide.
  • Lyxumia 10 micrograms solution for injection: each dose contains 10 micrograms of lixisenatide (50 micrograms per mL).
  • Lyxumia 20 micrograms solution for injection: each dose contains 20 micrograms of lixisenatide (100 micrograms per mL).
  • The other components are glycerol 85%, sodium acetate trihydrate, methionine, metacresol, hydrochloric acid (for pH adjustment), sodium hydroxide solution (for pH correction) and water for injections.

Description of Lyxumia and content of the pack
Lyxumia is a clear and colourless solution for injection (injection) inside a glass cartridge
inserted into a pre-filled pen.
Each green pen of Lyxumia 10 micrograms solution for injection contains 3 mL of solution,
which is equivalent to 14 doses of 10 micrograms. Pack of 1 pre-filled pen.
Each purple pen of Lyxumia 20 micrograms solution for injection contains 3 mL of solution,
which is equivalent to 14 doses of 20 micrograms. Packs of 1, 2 or 6 pre-filled pens.
Not all pack sizes may be available in your country.
A starter pack is also available for use during the first 28 days of
therapy. The starter pack contains one green pen of Lyxumia 10 micrograms
solution for injection and one purple pen of Lyxumia 20 micrograms solution for injection.

INSTRUCTIONS FOR USE
Each pre-filled pen contains 14 doses, each dose containing 10micrograms in 0.2 mL.

Section 1 - IMPORTANT INFORMATION
Read these instructions carefully before using the Lyxumia pen.

Keep this leaflet for future reference.

About the Lyxumia pen
Lyxumia is supplied in a pre-filled pen for injection.

  • Inject one dose daily.
  • Each Lyxumia pen contains 14 pre-set doses. You do not need to measure each dose.
  • Before using, ask your doctor, pharmacist or nurse how to inject correctly.
  • If you are unable to follow the instructions exactly or are unable to handle the pen (e.g. if you have vision problems), only use it with the help of someone who can assist you.

The Lyxumia pen

  • This pen must be used by one person only. It must not be shared with anyone.
  • Always check the label to make sure you have the correct Lyxumia pen. Also check the expiry date. Using the wrong medicine can be harmful to your health.
  • Do not try to draw the liquid out of the cartridge with a syringe.

The needle(supplied separately)

  • Only use needles approved for use with Lyxumia. Use 29 to 32 G single-use needles for the Lyxumia pen device. Ask your doctor, pharmacist or nurse which needle thickness and length are most suitable for you.
  • If the injection is carried out by another person, they must be careful not to accidentally injure anyone with the needle. There may be a risk of infection.
  • Always use a new needle for each injection. This helps to prevent contamination of Lyxumia or possible blockage of the needle.

Section 2 - HOW TO START

  • Activate the pen on the day of the first injection

First, activate the new pen

  • Before injecting a dose- before performing the injection, you must remove excess liquid from the new pen. This process is only carried out once and is called “activation”. The following steps 1 to 5 show you how to do it.
  • Activation ensures that the pen works correctly and that the dose of the first injection is correct.
  • Do not repeatthe activation process, or you will not be able to obtain 14 doses from the Lyxumia pen. The following images show how the activation opening on the pen’s injection button changes after activation.

The pen is activated and ready for injections. After activation, the opening remains white.

How to activate the new Lyxumia pen
Step 1 Remove the pen cap and check it

without particles. If not, do not use this pen.
Contact your doctor, pharmacist or nurse.
Verify that the activation

Remove the outer and inner caps of the needle. Keep the outer needle cap - you
will need it later to remove the needle.

Orient the needle towards a suitable container (such as a paper cup or tissue) to collect the
liquid so that you can dispose of it.

Press the injection button completely.You may feel or hear a “click”.

Hold the injection button down and count slowly to 5 to expel the last drops.

Step 5 The pen is now activated.
Do not reactivate this pen.

not need to replace the needle between activation and the first injection.

Section 3 - DAILY USE OF THE PEN
Follow the instructions in this section only when the activation opening is white.
Inject one dose daily.

Check the liquid. It must be clear and colourless, without particles. If not, do not use this
pen.
If there are air bubbles, see the “Questions and answers” section.
Check the number of doses in the pen. They are indicated by the position of the black plunger on the
dose scale.
Verify that the activation opening is white. If it is orange, go to Section 2.
Check the pen label to make sure you have the correct medicine.
Step B Insert a new needle and remove the needle caps

Grasp a fold of skin and insert the needle (see the “Injection sites” section to know where to inject the medicine).

Press the injection button completely.You may feel or hear a “click”.

Keep the injection button pressed and count slowly to 5to receive the full dose.
The dose has now been delivered. Remove the needle from the skin.
Step E Remove the needle and dispose of it after each injection

Place the outer needle cap on a flat surface. Insert the needle into the outer cap of the needle.
Put the outer cap back on the needle.

Grip the outer needle cap to grasp it and use it to unscrew the needle from the pen.
Ask your pharmacist how to dispose of the needle you no longer use.
Put the cap back on the pen.

Step F Repeat the steps described in Paragraph 3 for each injection.
Dispose of the pen 14 days after activation, even if it still contains medicine.

Storage
General information

  • Keep your Lyxumia pens in a safe place out of the sight and reach of children.
  • Protect your Lyxumia pens from dust and dirt.
  • Replace the cap on the pen after each use to protect it from light.
  • Do not use Lyxumia after the expiry date stated on the label and the carton. The expiry date refers to the last day of the month indicated.

Before activating the pen:

  • Store your unused Lyxumia pens in the refrigerator, between 2 °C and 8 °C.
  • Do not freeze Lyxumia pens and do not use Lyxumia if it has been frozen.
  • Allow the pen to return to room temperature before using it.

After activating the pen:

  • Once activated, store the Lyxumia pen below 30 °C. Do not freeze Lyxumia after activation.
  • Do not store the Lyxumia pen with the needle attached. Leaving the needle attached could cause contamination and possible air entry that could make the dose inaccurate.
  • Once activated, the Lyxumia pen can be used for a maximum of 14 days. Dispose of the Lyxumia pen after 14 days, even if it still contains medicine.

Disposal

  • Replace the cap on the pen before disposing of the Lyxumia pen.

Care

  • Handle the Lyxumia pen with care.
  • You can clean the outside of the Lyxumia pen with a damp cloth.
  • Do not immerse the Lyxumia pen in a liquid, wash it or wet it (to lubricate it) - it could be damaged.
  • If you think the Lyxumia pen may be damaged, do not use it. Do not try to repair the pen.

Lyxumia must be injected under the skin, in one of the areas indicated in blue in the figure above.
These areas are located on the thigh, on the abdomen or on the upper arm. Ask your doctor, pharmacist or nurse how to perform the injection correctly.

Questions and answers
What should I do if I forget to activate the Lyxumia pen or if I give an injection before activation?

If you have given an injection before activating the pen, do not try to remedy the situation by giving a second injection. Ask your doctor, pharmacist or nurse for advice on controlling blood sugar levels.

What should I do if there are air bubbles in the container?
The presence of air bubbles in the container is normal - they will not harm you. The dose will be correct and you can continue to follow the instructions. If you need help, contact your doctor, pharmacist or nurse.

What should I do if no liquid comes out during activation?
The needle may be blocked or not properly attached. Remove the needle from the pen, put a new one on and repeat only steps 4 and 5. If no liquid still comes out, the Lyxumia pen may be damaged. Do not use this pack of Lyxumia. Contact your doctor, pharmacist or nurse.

What should I do if it is difficult to press the injection button completely?

Read these instructions carefully before using the Lyxumia pen.

Keep this leaflet for future reference.

About the Lyxumia pen
Lyxumia is supplied in a pre-filled pen for injection.

  • Inject only one dose per day.
  • Each Lyxumia pen contains 14 pre-set doses. You do not need to measure each dose.
  • Before use, ask your doctor, pharmacist or nurse how to perform the injection correctly.
  • If you are unable to follow the instructions exactly yourself or are unable to handle the pen (e.g. if you have vision problems), use it only with the help of someone who can assist you.

The Lyxumia pen

  • This pen must be used by one person only. It must not be shared with anyone.
  • Always check the label to make sure you have the correct Lyxumia pen. Also check the expiry date. Using the wrong medicine can be harmful to your health.
  • Do not try to draw the liquid out of the cartridge with a syringe.

The needle(supplied separately)

  • Use only needles approved for use with Lyxumia. Use 29 to 32 G single-use needles for the pen device with the Lyxumia pen. Ask your doctor, pharmacist or nurse which needle gauge and length are most suitable for you.
  • If the injection is performed by another person, they must be careful not to accidentally injure someone with the needle. There may be a risk of infection.
  • Always use a new needle for each injection. This helps prevent contamination of Lyxumia or possible blockage of the needle.

Paragraph 2 - HOW TO START

  • Activate the pen on the day of the first injection

First, activate the new pen

  • Before injecting a dose- before performing the injection, you must remove excess liquid from the new pen. This process is done only once and is called “activation”. The following points 1 to 5 show you how to do it.
  • Activation ensures that the pen works correctly and that the dose of the first injection is correct.
  • Do not repeatthe activation process, or you will not be able to get 14 doses from the Lyxumia pen.

How to activate the new Lyxumia pen
Step 1 Remove the pen cap and check it

Check the liquid. It should be clear and colorless, with no particles. If not, do not use this pen.
Contact your doctor, pharmacist or nurse.
Verify that the activation opening is orange.
Step 2 Insert a needle and remove the needle caps

Align the needle with the pen. Hold it straight while screwing it onto the pen.

Store

Be careful not to injure yourself while the needle is exposed.
Remove the outer cap and the inner cap of the needle. Keep the outer needle cap - you will need it later to remove the needle.
Step 3 Pull out the injection button

Orient the needle towards a suitable container (such as a paper cup or tissue) to collect the liquid so you can dispose of it.

Press the injection button completely.You may feel a “click”.

Keep the injection button pressed and count slowly to 5 to expel the last drops.

Uso della penna viola da 20 microgrammi
Per iniettare una dose con la penna Lyxumia viola da 20 microgrammi, segua tutte le fasi riportate nel
Paragrafo 3. Ripeta le fasi del Paragrafo 3 per le sue iniezioni giornaliere finché la penna sarà esaurita.

Informazioni generali

  • Tenga le sue penne Lyxumia in un luogo sicuro fuori dalla vista e dalla portata dei bambini.
  • Protegga le sue penne Lyxumia dalla polvere e dalla sporcizia.
  • Rimetta il cappuccio sulla penna dopo ciascun utilizzo per proteggerla dalla luce.

Cura

  • Maneggi la penna Lyxumia con attenzione.
  • Può pulire l’esterno della penna Lyxumia con un panno inumidito.
  • Non immerga la penna Lyxumia in un liquido, non la lavi né la bagni (per lubrificarla) - potrebbe danneggiarsi.
  • Se ritiene che la penna Lyxumia possa essere danneggiata, non la usi. Ne prenda una nuova. Non cerchi di riparare la penna.

uno nuovo e ripeta solo le Fasi 4 e 5. Se continua a non uscire liquido, la penna Lyxumia potrebbe
essere danneggiata. Non usi questa confezione di inizio trattamento di Lyxumia. Contatti il medico, il
farmacista o l’infermiere.

Cosa devo fare se è difficile premere completamente il pulsante di iniezione?
L’ago potrebbe essere ostruito o non essere fissato correttamente. Estragga l’ago dalla pelle e lo tolga
dalla penna. Metta un ago nuovo e ripeta solo le Fasi D ed E. Se il pulsante di iniezione è ancora
difficile da premere, la penna Lyxumia potrebbe essere danneggiata. Non usi questa confezione di
inizio trattamento di Lyxumia. Contatti il medico, il farmacista o l’infermiere.
